What is Tris base?
Tris base is a white crystalline or powder. Soluble in ethanol and water, slightly soluble in ethyl acetate and benzene, insoluble in ether and carbon tetrachloride, corrosive to copper and aluminum, and irritating chemical substance.
Tris base is a sodium free amino buffer base that reacts with H2CO3 in body fluids, reducing H2CO3 and generating HCO32-. It can take up hydrogen ions and correct acidosis, with strong effects and can penetrate cell membranes. It is commonly used in acute metabolic and respiratory acidosis.
Tris base is a weak base with a pKa of 8.1 at 25 ℃; According to buffering theory, the effective buffering range of Tris buffer is between pH 7.0 and 9.2. The pH of Tris base aqueous solution is around 10.5. Generally, hydrochloric acid is added to adjust the pH value to the desired value, and a buffer solution with this pH value can be obtained. However, attention should also be paid to the effect of temperature on the pKa of Tris.
